摘 要: 白色链霉菌(Streptomyces albulus)出发菌株采用紫外诱变和硫酸二乙酯(DES)化学诱变进行复合诱变处理,处理液接种于甘氨酸和多聚赖氨酸抗性平板选育。通过优化甘氨酸和多聚赖氨酸剂量、紫外照射剂量和化学诱变剂DES剂量等诱变条件,并对诱变后的菌株进行初筛和复筛,最后得到一株较高产ε-多聚赖氨酸的菌株,ε-聚赖氨酸的产量达到0.73g/L的较高水平,是出发菌株的2.2倍。经5次传代发酵表明该菌株稳定。 Using Streptomyces albulus as starting strain, mutant strain with combined UV and diethyl sulfate(DES) was inoculated into plate with glycin and resistant ε-polylysine medium for screening of high ε-polylysine producing strain. The mutation conditions, such as concentration of Gly and ε-polylysine, dose of UV irradiation, dose of DES, were optimized, and then a high ε-polylysine producing strain was obtained after optimization of concentration of glycin and ε-polylysine, dosage of UV and diethyl sulfate (DES) and secondary screening. Production of ε-polylysine reached 0.73g/L, which was 2.2 fold of starting strain. The breeding mutant showed good passage stability in five generation cultivation.
