酿酒酵母S.cerevisiae YQ-7的高密度发酵
High density fermentation of Saccharomyces cerevisiae YQ-7

摘  要: 研究了酿酒酵母s.cerevisiae YQ-7发酵的最佳培养基组成;通过单因素试验优化碳源和氮源的添加量,响应面法优化培养基中的碳氮比,得出最佳发酵培养基配方:蔗糖76.31g/L,酵母浸膏33.75g/L,酸水解酪蛋白16.00gm,硫酸铵0.5班,硫酸镁1.OgrL,磷酸二氢钾2.6g/L,甘氨酸2g/L。同时,对补糖策略进行了研究,确定补糖工艺为自发酵16h开始补糖,补糖量70%~80%,每隔12h补糖一次,50h终止发酵,生物量为49.47ga,。 The optimal medium composition for Saccharomyces cerevisiae YQ-7 fermentation was studied. The addition amounts of carbon and nitrogen sources were optimized by single factor test, while the C/N ratio of the medium was optimized by response surface methodology. The results showed that the optimal formula of the fermentation medium were sucrose 76.31 g/L, yeast extract 33.75g/L, acid hydrolyzed casein 16.00g/L, (NH4)2SO4 0.5 g/L, MgSO4 1.0 g/L, KH2PO4 2.6 g/L and glycin 2 g/L. The sugar-feeding strategy was also investigated and determined as follows: the sugar-feeding started at 16 h of self-fermentation with 70%-80% sugar, and repeated at 28 h and 40 h. The biomass reached 49.47 g/L when the fermentation ended at 50 h.
