摘 要: 为了制备含高活菌量的LCR冻干粉,研究了增菌培养基和培养条件,并优化冻干保护剂。与MRS基础培养基相比,分别添加氮源、番茄汁、胡萝卜汁和抗坏血酸,LCR719活菌数分别提高了约2.0、1.3、1.2、1.5倍;以添加2.5倍氮源、8%番茄汁、6%胡萝卜汁、0.04%抗坏血酸的培养基,初始pH为6.8时,增菌效果最好,其活菌数达109cfu/mL,菌体最佳收获时间为16h;保护效果由大到小依次为海藻糖、脱脂奶粉、蔗糖、甘油、可溶性淀粉、硫酸锰、抗坏血酸;以10%海藻糖+10%脱脂奶粉的组合为最佳,其菌体存活率达100%,冻干菌粉活菌数达1011cfu/g。 In order to prepare the freeze-dried LCR 719 with high viable counts, the multiplication factors and culture conditions were investigated, and the cryoprotectances were optimized.in comparison with original MRS the viable counts of LCR 719 increase to 2.0,1.3, 1.2,1.5 times when the medium was added with nitrogen, tomato juice,carrot juice and Vc, respectively. The optimal conditions to multiply LCR 719 were as follows: 2.5 times nitrogen,8% tomato juice, 6% carrot juice,0.04% Vc into MRS, initial pH 6.8, the viable count of whose was 109cfu/mL,and the best time of capturing LCR 719 was 16h.The effect of seven cryoprotectances followed the order as that: D-Trehalose, Skim Milk, Sucrose, Glycerol, Soluble starch, MnSO4, Vc. The survivability rate was 100% when 10% D-Trehalose and 10% Skim milk were used into the freeze-drying of LCR 719.The viable count of the freeze-dried power was 10^11cfu/g.
