摘 要: 目的:检测高住低练训练方法和高糖高脂饮食对肾素血管紧张素系统(RAS)的基因表达的影响,从而提出治疗新血管疾病的新方法。方法:高住低练和高糖高脂饮食对大鼠心脏RAS基因表达的影响是通过real time PCR实现的,这种方法广泛被人们使用并且已经成为研究基因表达最为有效的方法。结果:我们的结果表明与安静组相比,高住低练和高糖高脂饮食可以显著降低ACE基因表达,其中高住低练组与高糖高脂高住低练组下调最多,血管紧张素原(AGT)的基因表达在低住低练组和高住安静组轻微上调,其他实验组都下调,其中高住低练组与高糖高脂高住低练组下调最多。与此相反的是,以上两种条件可以显著上调血管紧张素II的I型受体(AT1)基因的表达,高住低练组与高糖高脂高住低练组下调最多。结论:表明合理的训练方法(海拔2500m的高原环境下居住,适宜的训练强度)和高糖高脂饮食可以改善优秀运动员的有氧运动能力。同时高住低练的训练方法可以抑制高糖高脂饲料引起的胰岛素抵抗等症状。 Object: Rennin- angiotensin system(RAS) take an important role in regulating the blood pressure as well as water homeostasis. RAS gene expression is related with living high training low(IHTL) and high glucose high fat diet. So the aim of this study is to detect the effect of exercise training and diet on the Renin - angiotensin system gene expression. Method : The effect of IHTL and high - glucose - high - fat diet on RAS gene expression in the rat heart are investigated by real time RT - PCR which is the most powerfully methods for the research of gene expression. Result: Our result show that IHTL and high - glucose -high -fat diet can significantly decreased the gene expression of angiotensin- converting enzyme(ACE) and anglotensinogen (AGT). To be contrast, these two treatment can significantly increased the gene expression of angiotensinlI receptor type I (ATI). Conclusion: This study suggest that IHTL and high- glucose- high- fat diet can better improved the human's physical performance. Meanwhile, there may be a new therapy to hypertension and diabetes according to our result.
