摘 要: 本试验以番茄"中蔬四号"品种为试材,对不同激素组合、不同种类外植体、子叶不同部位及接种方式对番茄不定芽再生的影响进行了研究,同时还筛选了种子消毒的最佳方法和不定芽生根的最佳培养基。研究结果表明,番茄种子表面消毒宜采用70%乙醇浸泡30s,然后再用20%的次氯酸钠加土温消毒20min;子叶中间段为最佳外植体,茎段次之,真叶的诱芽率最低;子叶诱芽的最佳培养基为MS+ZT2mg/L+IAA0.1mg/L,且背面朝上放置更有利于不定芽的分化;不定芽生根的最佳培养基为1/2MS+IAA0.3mg/L。这项研究为番茄转化体系的建立奠定了基础。 In this research,we studied the regeneration of adventitious buds under the conditions of different hormone combination,different explants and parts of cotyledons and cotyledon inoculation method by employing "Zhong shu 4 hao"as experiment almaterial.Also,we screened the best method for seeds surface disinfection and the most appropriate medium forthe rooting of adventitious buds.The results showed that the best method forsurface disinfection of tomato seeds is as follows:soak the seed in 70% ethanol for 30 s firstly,then floated them in 20%NaClO and tween-20 for 20 min;the middle part of the cotyledon is the optimum explant,the stem took the second place,the leaf was the lowest;the best cotyledon regeneration medium was MS+ZT 2 mg/L+IAA 0.1 mg/L;placing the cotyledon upside down is more beneficial to induce the adventitious buds than upside;the most suitable medium forrooting of adventitious buds was 1/2MS+IAA 0.3 mg/L.This research would provide insights into establishment of transformation system of tomato.
