摘 要: 利用化学法合成甲基苯丙胺人工抗原,将此抗原免疫新西兰大白兔制备抗甲基苯丙胺多克隆抗体,利用EDC法将甲基苯丙胺人工抗原与蛋白质载体BSA偶联,经蛋白质电泳以及紫外扫描检测,证明甲基苯丙胺人工抗原合成成功.用间接酶联免疫检测法检测,证明制备得到了高效价的甲基苯丙胺多克隆抗体血清,且与甲基苯丙胺人工抗原具有高特异性反应,可用于甲基苯丙胺免疫学检测方法的研究。 Artificial methamphetamine antigen was chemically synthesized and was then used to immunize the New Zealand rabbits to prepare the polyclonal anti-methamphetamine antibody.Then the methamphetamine antigen was respectively coupled with BSA by means of EDC method.UV scan and protein electrophoresis showed that the artificial methamphetamine antigen was successfully synthesized.Moreover,ELISA results indicated that the rabbit serum with high dilute degree was prepared,and a high specificity reaction occurred between the polyclonal antibody and the antigen.Both the antigen and the antibody can be applied to the research on methamphetamine immunoassay.
