摘 要: 目的观察2,2′,4,4′-四溴联苯醚(BDE-47)暴露对小鼠肝脏甲状腺激素受体亚型TRα1、TRβ1在mRNA及蛋白水平表达的影响。方法将C57BL/6小鼠分成对照组和低、中、高三个染毒剂量组,对照组给予玉米油,染毒组分别按0.5、5和50mg/kgBW剂量给予BDE-47,连续4天腹腔注射后,以GAPDH为内参,实时荧光定量PCR(QRT-PCR)和蛋白印迹(Westernblot)测定肝脏组织甲状腺激素受体TRα1、TRβ1mRNA和蛋白表达。结果与对照组相比,中、高染毒剂量组小鼠肝脏TRα1和TRβ1mRNA表达均上调(P<0.05),幅度分别为1.9、1.7和1.9、1.5倍(P<0.05);三个剂量组TRα1蛋白表达均升高,TRβ1均下降。结论 BDE-47可改变小鼠肝脏TRα1、TRβ1mRNA和蛋白表达。 Objective To study the effect of 2,2′,4,4′-tetrabrominated diphenyl ethers (BDE-47) on the expression change of TRα1,TRβ1 at both mRNA and protein levels.Methods C57BL /6 mice were divided randomly into 4 groups and administrated with corn oil,0.5,5 and 50mg /kg /BW BDE-47 for 4 days through intraperitoneal injection respectively.GAPDH selected as internal standard,real time quantitative RT-PCR and Western blot were employed to detect the mRNA levels and protein of TRα1 and TRβ1 in livers.Results Compared with the control group,the mRNA levels of TRα1 and TRβ1 were up-regulated in the medium and high BDE-47 dose groups(P〈0.05).The protein level of TRα1 significantly up-regulated while TRβ1 was significantly downregulated.Conclusion BDE-47 could change the expression of TRα1 and TRβ1 in the mRNA and protein levels in liver.
