摘 要: 以陇东野生紫花苜蓿、阿尔冈金、甘农1号、游客4个苜蓿品种的下胚轴为外植体,比较不同苜蓿品种外植体愈伤组织诱导、分化及再生能力的差异,研究不同激素浓度及配比对再生体系建立的影响。结果表明,4个苜蓿品种下胚轴诱导率依次为阿尔冈金〉甘农1号〉陇东野生紫花苜蓿〉游客;在愈伤组织诱导培养基MS+2.0 mg/L 2,4-D+1.0 mg/L NAA上诱导率最高;MS+0.5 mg/L KT+0.3 mg/L NAA为最佳分化培养基,分化率在84.5%~93.5%;适宜的生根培养基为1/2 MS+0.1 mg/L NAA,附加1%蔗糖,各品种再生植株的移栽成活率为100%。 Effects of different hormone concentrations and ratios on establishment of regeneration system of alfalfa was detected baed on callus induction,differentiation and regeneration abilities of the four alfalfa varieties with its hypocotyls as explants.The results indicated that the highest inducement ratio of hypocotyls was Longdong Medicago sativa,followed by Algonquin,Gannong No.1 and Eureka.The best callus induction medium was MS + 2.0 mg/L2,4-D + 1.0 mg/LNAA,while MS + 0.5 mg/L KT + 0.3 mg/LNAA was the best differentiation medium,and its differentiation ratio was 84.5%~93.5%.The best shooting medium was 1/2MS + 0.1mg/LNAA with 1% sucrose.The survival rate of transplanting of the above varieties was 100%.
