一个新型嗜热菌质粒的测序鉴定及抗生素抗性研究

摘  要: 一株分离自80℃温泉的嗜热菌对卡那霉素表现出抗性,推测这株菌可能携带有卡那霉素抗性的质粒.质粒消除后的抗性试验和质粒转化到大肠杆菌JM109中的抗性试验都表明该质粒具有卡那霉素抗性.将该质粒部分序列克隆测序后与Thermus属2个质粒的同源性最高达到89%,这进一步证实了这株嗜热菌里确实有嗜热质粒存在. A thermophilic bacteria isolated from the hot spring with 80℃ shows the antibiotic resistance to kanamycin.It is speculated that the strain is likely to take plasmid with the antibiotic resistance.The antibiotic resistance experiments after plasmid was eliminated and of plasmid was transformed to E.coli JM109 showed that the plasmid has the antibiotic resistance to kanamycin.After its partial sequence was cloned and sequenced.The result of nucleic acid sequence shows that the homology of the partial sequence of the plasmid and that of two submitted plasmid sequences in Thermus genus in GenBank was up to 89%.It was favored further that there is a thermophilic plasmid in the thermophilic bacteria.
