摘　　要： 目的:研究酵母表达中性粒细胞明胶酶相关脂质运载蛋白(NGAL)的纯化方法。方法:表达上清用(NH4)2SO4盐析、凝胶过滤、阳、阴离子交换层析等方式进行纯化。通过比较产品纯度、收率与质量确定最佳实验方案。纯化产品用SDS-聚丙烯酰胺凝胶电泳、Western blot、等电聚焦与高效液相色谱进行鉴定。结果:获得较高纯度的NGAL。结论:(NH4)2SO4盐析、凝胶过滤和阳离子交换是纯化酵母表达NGAL的最佳实验方案。 Objective：To study the purification method of neutrophil gelatinase-associated lipocalin（NGAL）expressed in yeast.Methods：The protein was purified by ammonium sulfate precipitation,gel filtration,cation and anion exchange chromatography.The optimum purification plan was determined by comparing the product purity,yield and quality.The products were identified by SDS-polyacrylamide gel electrophoresis,Western blot,isoelectric focusing and high performance liquid chromatography.Results：The high quality NGAL was obtained.Conclusion：The mmonium sulfate precipitation,gel filtration and cation exchange are the best choice for purification of NGAL expressed in yeast.
