摘 要: 目的:探讨胼胝体内主要蛋白的构成。方法:通过二维电泳技术分离小鼠胼胝体的蛋白,用PDQuest软件对蛋白点进行分析,用串联飞行质谱仪对蛋白进行鉴定,并用Western Blot和免疫组织化学对鉴定的蛋白进行分析。结果:二维凝胶电泳显示在小鼠胼胝体有679个蛋白点。选取47个高丰度蛋白点进行质谱鉴定,鉴定为44种蛋白,它们分别为结构、代谢、信号转导、转运和应激蛋白。WesternBlot和免疫组织化学方法进一步证实了蛋白在胼胝体的表达和分布。结论:该结果为胼胝体主要蛋白的构成提供了资料。 Objective: To identify high abundant proteins in the corpus callosum. Methods: Proteins of the corpus callosum were separated by two-dimensional (2-D) gel electrophoresis, and analysed using PDQuest software. Proteins on the 2-D image were identified through matrix-assisted laser desorption/ionization two-stage time of flight ( MALDI TOF / TOF). We further investigated the expression and distribution of some specific proteins in the corpus callosum using Western blot and immunohistochemistry. Results: 2-D image showed that there were 679 protein spots in the corpus callosum. 47 high abundant protein spots were chosen to be analysed. They were identified as 44 proteins, which were involved in the cytoskeleton, metabolism, signaling pathway, transport and stress response. Our study further confirmed expression and distribution of proteins in the corpus eollosum by western blot and immunohistochemical methods. Conclusion: Our results provided data for high abundant proteins in the corpus callosnm.
