利用ISSR技术检测梅菜试管苗的遗传稳定性研究

摘  要: 利用植物组织培养方法建立起梅菜无性系,并连续进行5次继代培养;利用已优化的ISSR-PCR反应体系对不同继代次数(1~5代)的梅菜试管苗进行遗传稳定性分析。结果表明:在梅菜无性系继代过程中,从试管苗的形态方面观测没有发生明显的变化。但利用已优化的ISSR-PCR检测技术对2个梅菜品种亲本及不同继代试管苗进行检测,2个品种之间带型差异明显,同1个品种在不同世代带型基本一致,但从第4代开始,部分引物带型发生变化,但变化甚微,具有较高的遗传稳定性,在4代以内仍可稳定遗传。 The aim was to dectct the genetic stability in tube seedlings of Brassica juncea Coss. var. sp nov from the molec- ular level by ISSR molecular markers, which provided the theory basis for using the Brassica juncea Coss. vat. sp nov. The Plant culture tissue method was used to establish the Brassica juncea Coss. vat. sp nov clone, and was subcultured culture 5 times continuously; the ISSR-PCR reacting system which optimized already to analyze the hereditary stability of tube seedlings with different subcultured times(1-5 generations). The results indicated that in the suhcultured culture process of Brassica juncea Coss. vat. sp nov clone, there was no obvious changes were observed from the tube seedling's shape. But used ISSR-PCR examination technology to assay two varieties parent and tube seedlings with different subcultured times, between two varieties the band difference was obvious, and same varieties in different generation was basic consistent, but the slight variations of some primer bands were detected since the 4th descends, which was only the change of the clearance of the band. The genetic stability of Brassica juncea Coss. vat. sp nov repeatedly subcultured in vitro with the material of callus was maintained within four descends, though some variations were found. This research provided the theory basis for the plum vegetable clone selective breeding and the idioplasm appraisal.
