大白菜游离小孢子胚诱导及植株再生
Isolated Microspore Culture and Plantlet Formation in Heading Chinese Cabbage

摘  要: 以15个大白菜[Brassica campestris ssp. Pekinensis (Lour.) Olsson]自交系和杂交种为试材进行游离小孢子培养,研究了影响胚状体发生及植株再生的几个主要因素。结果表明,有12个品种的游离小孢子在NLN-13液体培养基上诱导出胚;各品种间小孢子胚产量差别很大,每个花蕾产胚在0.25~65.4个之间;将成熟的子叶形胚及时转移至再生培养基上,对提高植株获得率至关重要;在NLN-13液体培养基上附加0.2mg.L-1的6-BA对胚状体的发生和发育有促进作用;B5+0.2 mg.L-16-BA+0.02 mg.L-1NAA的培养基有利于小孢子胚发育成植株;1/2 MS+0.1 mg.L-1NAA为小孢子植株生根最适宜的培养基。 Isolated microspore culture was carried out with 15 F1 hybrids and inbred lines of heading Chinese cabbage(Brassica campestris ssp.Pekinensis(Lour.) Olsson).Major factors influenced embryogenesis in Chinese cabbage during microspore culture were investigated.The results showed that 12 varieties of heading Chinese cabbages were cultured in NLN-13 liquid medium and produced microspore-derived embryos and plantlet regeneration.The production of microspore-derived embryos varied remarkably among these varieties.Embryo yield per bud ranged from 0.25 to 65.4.Transferring mature cotyledonary embryos to the regeneration medium in time was essential to increase plantlet regeneration rate.The effect of different hormone concentrations in NLN medium on embryogenesis was compared.It showed that NLN-13 medium added with 0.2 mg·L-1 6-BA promoted genesis and development of the embryoids.Microspore-derived embryos on B5 medium with 6-BA 0.2 mg·L-1and NAA 0.02 mg· L-1 were favorable to regenerating plantlets.For rooting of microspore plantlets,1∕2 MS added with 0.1 mg·L-1 NAA was suitable.
