摘 要: 目的比较琼脂糖平铺法与poly-HEMA包被法悬浮培养人骨肉瘤细胞建立人抗失巢凋亡骨肉瘤细胞模型的异同和优劣。方法选择hFOB1.19恶性转化细胞和SaOS-2骨肉瘤细胞,分别采用琼脂糖平铺法和poly-HEMA包被法进行悬浮培养建立人抗失巢凋亡骨肉瘤细胞模型,倒置相差显微镜、透射电子显微镜观察悬浮培养后细胞的形态学改变。结果琼脂糖平铺法悬浮培养人骨肉瘤细胞可成功建立人抗失巢凋亡骨肉瘤细胞模型,且与poly-HEMA包被法相比较,琼脂糖平铺法价格经济、操作容易、效果更优。结论琼脂糖平铺法悬浮培养细胞建立抗失巢凋亡细胞模型更适用。 Objective To compare features,advantages and disadvantages of two anti-adhesion cell culture methods by covering agarose and coating poly-HEMA on cell culture plates in establishing human anoikis-resistant osteosarcoma cell models,respectively.Methods A malignant transformed hFOB1.19 cell lines and SaOS-2 osteosarcoma cell lines were cultured in the cell culture plates covered with a layer of agarose and poly-HEMA,respectively.Inverted phase contrast microscopy and transmission electron microscopy were used to observe their morphological changes.Results A human anoikis-resistant osteosarcoma cell model had been established successfully by anti-adhesion cell culture method by covering a layer of agarose on cell culture plate.And this method was economical,easy-operating,and effective,comparing with the one by coating poly-HEMA.Conclusions The anti-adhesion cell culture method by covering agarose on cell culture plates is more applicable than by coating poly-HEMA in establishing human anoikis-resistant osteosarcoma cell models.
