摘 要: 对经碱性蛋白酶Alcalase 2.4L水解的牛骨胶原蛋白肽清除自由基活性进行了研究。根据对羟基自由基、超氧阴离子自由基、二苯代苦味酰基自由清除能力的测定,以及还原能力、脂质抗氧化能力等抗氧化活性指标的测定,确定了水解度为7.9%左右的胶原蛋白肽清除自由基的综合效果最好,此时酶解产物的平均分子量为2025Da,含15个左右的氨基酸。在此条件下,超氧阴离子(O2-·)的清除率为63.86%,羟基自由基(·OH)的清除率为99.55%,二苯代苦味酰基自由基(DPPH·)的清除率为27.03%,对大豆卵磷脂过氧化作用的抑制率为74.78%。 Enzymatic hydrolysis of collagen from ox bone, using Alcalase 2.4L as biocatalyst, was studied to scavenge activity free radicals.Considering the total scavenging activity of free radicals of hydrolysis product, the best collagen peptide was prepared by controlling the reaction DH of about 7.9% (reaction time 30min).ln this situation, ·OH scavenging value was 99.55%, O2^-· scavenging value was 63.86%, DPPH · scavenging value was 27.03% ,the anti-lipid peroxidation activity of the peptide was 74.78%. Collagen peptide with highest total anti-radicals activity contained 14-15 amino acids with its average molar weight of 2025Da.
