摘 要: 对超声波辅助提取芸豆种皮水溶性多糖的优化工艺进行研究。在将芸豆种皮进行乙醇回流处理后,根据中心组合设计原理,在单因素基础上,选定pH、液料比、超声波功率和超声提取时间为实验因素,以芸豆皮水溶性多糖得率为目标建立回归数学模型,通过实验结果进行方差分析及对数学模型进行优化得到芸豆皮水溶性多糖的优化提取条件。实验结果表明,当pH为4.63、液料比为20∶1(mL∶g)、超声波功率为238W、超声提取时间为59min时,芸豆种皮水溶性多糖的理论得率为10.38%,验证值为10.43%。 The extraction technology of water-soluble polysaccharide from kidney bean hull by ultrasonic-assisted was studied ,and response surface design was applied to optimize the extraction technology.On the basis of single -factors experiments,the center-composite experimental design principles were used to design the experiments and the experimental factors of pH value,the rate of liquid to solid,ultrasonic power and ultrasonic extraction time were adopted after the kidney bean hull treated with ethanol.The regression mathematical model for yield of watersoluble polysaccharide of kidney bean hull was established,and the optimum extraction condition was obtained by variance analysis and the optimization of mathematical model. Results showed that the optimum extraction conditions were obtained as follows: pH value of 4.63,the rate of liquid to solid ( mL: g ) of 20:1, ultrasonic power of 238W and ultrasonic extraction time of 59min, under which the highest yield of watersoluble polysaccharide of kidney bean hull reached with estimated value and verified value being of 10.38% and 10.43% ,respectively.
