摘 要: 利用超速离心纯化的鸡传染性支气管炎病毒(IBV)广东地方分离株D41免疫BALB/c小鼠。取脾细胞与SP2/0骨髓瘤细胞进行融合,经筛选及克隆,成功地建立了6株可分泌抗IBVD41株的单克隆抗体(McAb)的杂交瘤细胞:B5、B8、C7、D8、D9、G11。其中B8、C7、G11的稳定性最好,腹水McAb效价高达1010,无血清培养上清McAb(浓缩30倍)效价达4×104以上。6株杂交瘤细胞的特异性均很好。 s In this study,avian infectious bronchitis virus(IBV)strain D 41 isolated from Guangdong province was used for antigen and purified by sucrose density gradient centrifugation. The monoclonal antibodies(McAbs)were generated by fusing Sp2/0 mouse myeloma cells and splenocytes from BALB/c mice immunized with the antigen. After screening and cloning,six murine hybridoma cell strains (B 5、B 8、C 7、D 8、D 9、G 11 )which secrete specific McAbs to IBV strain D 41 were successfully established. Three of the six hybridomas(B 8、C 7、G 11 )are more stable.Their ascitic fluids titers and serum-free culture fluids(condensed 30 times titers were more than 10 10 ×10 4 respectively.
