摘 要: 基于防腐防霉剂构效关系研究,以顺丁烯二酸酐、丙三醇和甲醇为原料,合成一种具有α,β-不饱和羰基结构的新型食品防腐剂——富马酸甘油甲酯.用单因素法获得的较优合成条件为:以占反应物总质量6%的浓硫酸为催化剂,按醇酐摩尔比1∶1加入顺丁烯二酸酐和丙三醇,在60℃下酯化反应4 h,得到顺丁烯二酸甘油单酯,以占反应物总质量4%的无水AlCl3为异构化催化剂于90℃下异构化2 h,得到富马酸丙三醇酯,再加入醇酐摩尔比1.2∶1.0的甲醇在60℃下酯化5 h,即可得到产品富马酸甘油甲酯,产率为73.5%.文中对其抑菌活性作了考察,结果表明:富马酸甘油甲酯的抗菌活性强于常用防腐剂苯甲酸和山梨酸钾,具有广泛的开发应用前景. Based on the structure-activity relationship of preservative and mold preventives, a new type of preservatives named glycerine methyl-fumarate (GMF) was synthesized from maleic anhydride, glycerine, and methanol. Also, its antimicrobial activity was studied. The factors affecting the yield were investigated by mono-factor test. Firstly, the glycerine maleate was synthesised with mole ratio of glycerine and maleic anhydride 1.0 : 1.0, and H2SO4 with 6% mass fraction as catalyst, esterification for 4h at 60℃. Secondly, glycerine fumarate was synthesised by AlCl3 with 3% mass fraction as isomerization catalyst, isomerizationabput for 2h at 80℃. Thirdly, glycerine methyl fumarate was synthesised with mole ratio of the methanol and the maleic anhydride 1.5 : 1.0, esterification for 3h at 60℃. Under the above conditions, the yield of glycerine methyl-fumarate was over 73.5 %. The antibacterial experiments show that its antibacterial activity is much better than these of benzoic acid and potassium sorbate, which means its development and application are wide.
