摘 要: 为探讨免疫组织化学Envision法在诊断猪繁殖与呼吸综合征(PRRS)中应用的可行性,采用免疫组织化学Envision二步法对30例疑似PRRS病猪肺组织的病毒抗原进行定位、半定量检测。结果表明,Envision法可原位检测病猪肺组织PRRSV抗原的分布;其抗原的阳性表达主要出现在肺巨噬细胞胞浆内,其次为肺泡上皮细胞和细支气管黏膜上皮细胞。依据阳性细胞百分率,进行染色结果判断,检出阳性病猪23例,阳性检出率为76.7%。试验证明,Envision法具有高敏感、低背景、快速简便的特点,可在PRRS诊断中推广应用。 To observe the value of Envision method of immunohistochemistry techniques for detecting of porcine reproductive and respiratory syndrome (PRRS),the antigens of PRRSV were detected in-situ and semiquantitative by Envision method in lung tissues of 30 suspected PRRS swine. The results showed the PRRSV antigens could be located by Envision method. Positive express of antigens was mostly in the cytoplasm of porcine alveolar macrophages, and was posteriorly in alveolar and bronchiole epithelia. According to the positive cells percentage of Envision staining,23 samples were positive, positive rate were 76.7 %. Envision method with highly sensitive, low background, rapid and convenient features,can be applied to detecting of PRRS.
