摘 要: 目的:分析市售鸡蛋的蛋白图谱,找出共性,建立鸡蛋稳定的蛋白质模式图谱,作为真假鸡蛋鉴别的检测方法。方法:对鸡蛋蛋黄的蛋白质进行等电聚焦(pH4~7)、SDS-PAGE电泳,银染后扫描,对获得的双向图谱用软件进行分析。结果:根据18个市售鸡蛋与真鸡蛋的蛋黄蛋白质图谱对比,获得三个共有的高丰度点群(共17个蛋白质点),分子量分别为97.2~66.4、66.4~44.3、44.3~29.0kDa,可作为鉴别真假鸡蛋的蛋白质模式图谱。市售鸡蛋蛋黄的蛋白质图谱与真鸡蛋相似,没有发现异样情况,没有在市场上发现假鸡蛋。结论:初步建立了鉴别真假鸡蛋的蛋白质组学方法,所建立的检测方法具有高度的重复性和准确性。 Objective: To establish a detection method for distinguishing real eggs from man-made eggs by finding the common model proteins map through two-dimensional electrophoresis. Method: Acquired the egg yolk' s proteins maps by isoelectric focusing, SDS-PAGE and silver stain. The maps then had been scanned into computer and analyzed by software. Result: Compared proteins maps of 18 market-sold egg samples to that of the two real hen eggs, showed that there were three mutual abundant protein spots groups ( 17 protein spots in total) ,which molecular weights were 97.2-66.4kDa,66.4-44.3kDa,44.3-29.0kDa respectively and could be used to construct the model proteins map in detecting real eggs or not.All sample' s protein maps are similar to that of the two real eggs,which showed that man-made eggs were not found in the markets.Conclusion: A detection method for distinguishing real eggs from man-made eggs by proteomics have been established, which was highly repeatable and accurate.
