阳离子交换树脂固定化乳糖酶的研究

摘  要: [目的]以阳离子交换树脂为载体,研究黑曲霉来源乳糖酶固定化条件,以期改善乳糖酶性质,使黑曲霉乳糖酶更适于低乳糖乳的连续生产。[方法]以吸附率最高的阳离子交换树脂D151为载体,通过先吸附后交联的方法固定乳糖酶,优化固定化条件。[结果]结果表明,加酶量为50.0 U/g(载体),吸附pH值为4.0,吸附温度是25℃,吸附时间24 h,交联剂戊二醛浓度为4%,交联温度是30℃,交联时间是6 h,固定化效果最好。获得的固定化酶活力可达11.8 U/g(载体),固定酶回收率为37.2%。[结论]该研究为工业化利用固定化乳糖酶连续生产低乳糖乳提供了技术依据。 [ Objective] The cation exchange resin being taken as the carrier, the immobilization condition of the lactase derived from aspergillus niger was researched so that the lactase from aspergillus niger was more suitable for the continuous production of low-lactose milk through the improvement of the property of lactase. [ Method] The cation exchange resin-D151 with the highest absorption rate being taken as the eartier, the immobilization condition was optimized through the experiment in the method of cross-linking after adsorption in the immobilization of laetase. [ Result] The results showed that the best immobilized efficiency condition were the quantity of enzyme(as carrier), 50.0 U/g; the pH value in adsorptionwas 4.0 ; the temperature was 25 ℃ ; the time was 24 hours ; and the concentration of glutaraldehyde ( as cross-linking agent) was 4% ; the temperature in cross-linking action was 30 ℃, the time was 6 hours. The activity of immobilized enzyme was up to 11.8 U/g, the recovery of enzyme fixed was 37.2%. [ Conclusion] The technical basis of the continuous production of low-lactose milk for industrial use of immobilized lactase was provided.
