乳脂链球菌产胞外多糖的发酵条件

摘  要: 通过单因素及正交实验对乳脂链球菌产胞外多糖的发酵条件进行研究。单因素实验研究了碳源、接种量、起始pH、发酵温度、镁元素和发酵时间对乳脂链球菌胞外多糖产量的影响,通过正交实验设计研究三个重要因素乳糖添加量、pH和接种量对胞外多糖产量的影响。优化处理后的最佳发酵条件是,发酵液中乳糖添加量为20 g/L,发酵培养基初始pH5.5,接种量5.0%,发酵温度为37℃,发酵时间24 h。胞外多糖最高产量可达499.81 mg/L。 Fermentation technology of EPS produced by Streptococcus lactis was studied through single factor and rthogonal experiment.During single factor test, the influence of different factors on EPS yield was analy- sized as different carbon sources, different lactose contents, different initial pH, different cultivation tempera- ture, different mineral quantity, and different fermentation time. Orthogonal experiment was carried by combin- ing the three factors, lactose contents, initial pH and inoculums under three different levels. The best fermenta- tion technology was obtained as lactose content 20 g/L, initial cultivation pHS.5, inoculum quantity 5.0%, fer- mentation temperature 37 ℃, fermentation time 24 h. The most yield of EPS is 499.81 mg/L.
