与冬瓜枯萎病抗性连锁的RAMP标记的筛选及其运用

摘  要: 枯萎病是制约冬瓜生产的主要因素之一,选育和种植抗病品种是控制冬瓜枯萎病的经济有效措施。本研究以冬瓜抗病材料B94与高感病材料B43的杂交F2后代分离群体为供试材料,利用RAMP技术,获得了与冬瓜枯萎病抗性连锁的RAPM分子标记CGA-6380。Mapmarker3.0软件分析CGA-6380与抗性基因的遗传距离为5.2cM。利用此标记对现有42份冬瓜种质资源的抗性进行了检测,并对17个杂交组合的抗性进行杂种优势分析和预测及对含B94抗性材料进行辅助回交育种,同时对这些材料进行了人工接种抗性鉴定。实验初步结果表明,42份材料中有12份达到抗性水平,17份与B94有亲缘关系,其中既与B94有亲缘关系,又具有抗性的材料有6份,说明CGA-6380适宜筛选携带有B94抗性基因的材料,含有CGA-6380条带的材料肯定有抗性,而没有CGA-6380条带的材料未必没有抗性。其它分子鉴定结果与人工鉴定结果基本一致。此研究结果将大大提高育种效率和进程,从而为选育冬瓜新种质和新品种提供参考。 Blight is one of the major constraint factors on yield loss in white gourd (Benincasa hispida (Thunb.) Cogn.) production. Breeding and cultivating white gourd cultivar with high resistance might be the most economical and efficienct way to control the blight disease in white gourd production. In this research, we constructed white gourd F2 populations derived from the cross of B94 and B43, of which B94 is a disease-resistant variety and B43 is highly susceptible as the parents, and then we obtained a RAPM marker CGA-6 380 by using the RAMP technique, which obviously close linked to resistance gene with 5.2 cM genetic distance based on the calculations of the software Mapmaker version 3.0. Furthermore, We used the acquired marker to validate 42 resistant germplasms, and to analyze and forecast 17 resistant combinations as well as to assist the selection of backcross breeding, meanwhile, artificial inoculation to identify the resistance of these tested materials was performed in this research. The preliminary results demostrated that 12 germplasms of 42 tested materials were displayed high resistance and 17 of them had well relationship with the parent B94, interestingly, 6 of tested materials not only had good relationship with B94, but also showed high resistance. The results indicated that CGA-6 380 should be suitable to validate the germplasm harboring B94 resistance gene, and then we may have a conclusion that the tested materials would have resistant phenotype to the white gourd blight if CGA-6 380 specific fragment could be validated, and we could not determine whether or not there is resistant chracters if no CGA-6 380 fragment amplified. The Results based on molecular approaches were quite similar as that of artificious identification. Therefore, we believed that this study might greatly increase the breeding efficiency and shorten the breeding periods and also it would provide the reference for developing the innovated germplasms in white gourd breeding program.
