不同培养基及ZT诱导对洋桔梗玻璃化的影响

摘  要: 以洋桔梗叶片为材料,以培养基MS+6-BA 0.5mg/L+IBA 0.2mg/L为对照,将在对照培养基上培养的叶盘转接到含不同激素组合(6-BA 0.2~0.5mg/L、IBA 0.05~0.1mg/L、NAA 0.01~0.2mg/L、ZT 0.5~1.0mg/L)的培养基中培养30d;将不同分化时期的愈伤组织每隔3d转接到MS培养基中培养30d,观察洋桔梗不定芽分化情况。结果表明,将已诱导15d的叶盘转接到MS+6-BA 0.3mg/L、MS+6-BA 0.4mg/L+IBA 0.05mg/L培养基中培养,利于正常不定芽的生长,分化总芽数和正常芽总数、平均每叶盘正常芽数明显高于对照,正常苗率与对照相差不明显。此外,将现芽点6d以前的不同分化程度的愈伤组织接到MS培养基中,则不利于正常不定芽的诱导分化;将现芽点第6d的愈伤组织转接到MS培养基中继续培养,其总分化芽数、正常芽总数、平均每叶盘正常分化芽数均高于对照而正常苗比率与对照相差不明显,说明现愈伤后不定芽的分化还继续需要外源激素的参与,以减少不定芽的玻璃化。 The present study has been conducted to study the effect of different media and hormone composition on the differentiation of adventitious buds in Enstoma grandiflorun The leaf pieces cultured in medium MS+6-BA 0.5 mg/L+IBA 0.2 mg/L(control medium) for 15 days were transferred to different media with different hormone composition(6-BA 0.2-0.5 mg/L; IBA 0.05-0.1 mg/L; NAA 0.01-0.2 mg/L; ZT 0.5-1.0 mg/L)for 30 days. The differentiated callus induced in control medium were transferred to MS medium every 3^rd day and cultured for 30 days to obverse the differentiation of bud. The results showed that two media MS+6-BA 0.3 mg/L and MS+6-BA 0.4 mg/L +IBA 0.05mg/L were beneficial to the growth of adventitious bud. The total differentiated buds, total normal buds and avemge normal buds per leaf piece were found to be higher compared to control, while no significant difference was observed in rate of normal buds production. When the 6 days callus with buds were transferred to MS medium, the induction in total differentiated buds, total normal buds and avemge normal buds per leaf piece were found to be higher than that of the control, while no signif- icant difference in rate of normal bud production was observed. Data revealed that the differentiation of buds induced from callus requires the exogenous hormone to reduce the vitrification.
