摘 要: 通过静脉注射的方法研究了大肠杆菌内毒素(ET)对山羊红细胞膜和肝粒体膜损伤的膜Ca2+-ATP酶活性的变化及654-2(山莨菪碱)的保护效应.结果表明,ET处理组(Ⅱ组)在1h,3h红细胞膜Ca2+-ATP酶活性高于对照组(Ⅰ组)(p<005),5h有下降趋势且持续到9h.12h开始回升并低于Ⅰ组(p<001,p<005).654-2处理组(Ⅲ组),Ca2+-ATP酶活性除在9h、12h与Ⅰ组,在3h与Ⅱ组差异不显著(p>005)外,均高于Ⅰ组和Ⅱ组(p<001,p<005).在肝线粒体中,Ⅱ组Ca2+-ATP酶活性在5h、12h均低于Ⅰ组和Ⅱ组(p<005,p<001),Ⅲ组Ca2+-ATP酶活性均高于Ⅰ组,但12h与Ⅰ组差异不显著(p>005).结果提示,ET具有诱导膜Ca2+-ATP酶活性先激活后顿抑的效应,而654-2有明显保护膜Ca2+-ATP酶活性的作用. The alterations of membrane Ca 2+ -ATPase activities of erythrocyte, hepatic mitochondria and protection of anisodamine(654-2) in goats were studied by intravenous injection of E. Coli (O 111 B 4) endotoxin (1 800 EU/kg body weight) and anisodamine (2 5 mg/kg body weight) alone as well as in combination. The results showed that endotoxin treatment (group Ⅱ)led to a marked increase of Ca 2+ -ATPase activities of erythrocyte membrane at 1 h and 3 h, a significant decrease of Ca 2+ -ATPase activities of erythrocyte membrane at 5 h, 7 h, 9 h, 12 h and liver mitochondria membrane at 5 h, 12 h in vivo, as compared to the normal control (group Ⅰ). Ca 2+ -ATPase activities in endotoxin and anisodamine coexposure group (group Ⅲ) were higher than those of group Ⅰand group Ⅱ, except no significant difference at 9 h, 12 h in erythrocyte membrane and at 12 h in liver mitochondria membrane as compared to grooup Ⅰduring all experiment. These results indicated that endotoxin could activate the membrane Ca 2+ -ATPase activities in the primary stage of administration of the endotoxin. Soon afterwards, Ca 2+ -ATPase activities were inhibited, whereas anisodamine had a prominent effect of protection on the Ca 2+ -ATPase of erythrocyte and hepatic mitochondria membrane of goats in vivo.
