摘 要: 为提高抗克伦特罗(clenbuterol,CBL)单链抗体(single chian Fv,scFv)的表达量,考察了Mg2+、Ca2+、NH4+、NaHPO4-等无机离子,初始pH值以及装液量对重组抗体表达量的影响。优化试验结果表明,重组大肠杆菌E.coli BL21在初始pH值为7.2、含2 mmol/L Ca2+和20 mmol/L NaHPO4-的培养基中培养,装液量为25 mL,42℃诱导表达后抗CBL-scFv的表达量为33.7%。 The fermentation process of recombinant anti-clenbutero (CBL) single chain Fv (scFv) was studied in order to improve the expression level of recombinant protein. The effects of factors such as, concentration of inorganicions, i. e. Mg^2+ , Ca^2+, NH4^+ and NaHPO4^-, pH ,and volume of medium were investigated. The results showed that the expression level of anticlenbuterol scFv was increased to about 33.7% of the total bacteria protein when recombinant E.coli was induced at 42℃ with pH 7.2, 25 mL volume of medium with 2 mmol/L Ca^2+ and 20 mmmol/L NaHPO4^-.
