摘 要: 以广东省5个家鹅品种36个样本为材料,对其线粒体细胞色素b基因的1 316 bp序列片段进行测序分析,研究5个家鹅品种的遗传多态性和起源。结果显示,在所有样本中,A、G、C、T碱基平均含量分别为28.84%、13.77%、34.23%、23.15%;共发现33个变异位点,变异类型为转换和颠换,未发现缺失与插入;确定了24种单倍型,H1为家鹅的主体单倍型,平均单倍型多样度为0.968,平均核苷酸多样度为0.01090;4个品种之间双参数遗传距离为0.0000~0.00146,其中马岗鹅与狮头鹅之间遗传距离最小,白沙鹅和马岗鹅、狮头鹅的遗传距离最大。结合NCBI上已发表的17个家鹅同源序列进行分析,共发现26种单倍型,单倍型聚类结果表明5个广东家鹅品种单一起源于鸿雁。 Thirty-six samples from 5 domestic goose breeds in Guangdong province were used as materials in this study. A 1 316 bp fragment of Cytochrome b gene in mitochondrial DNA was sequenced to analyze the genetic diversity and evolution of these goose breeds. Results showed that the A, G, C, T percentage in all samples were 28.84%, 13.77%, 34.23% and 23.15%, respectively. A total of 33 variation sites were found in 1 316 bp region, all of which were transitions and transversions. Twenty- four haplotypes were present in all samples, in which H1 was the major haplotype and the average nucleotide diversity (P/) and haplotype diversity (Hd) were 0.01090 and 0.968 each. The Kimura 2 parameter genetic distance was 0.0000-0.00146 within 5 domestic goose breeds, with the lowest distance between Magang goose and Shitou goose, and the highest distance between Baisha goose and Magang goose, as well as Baisha goose and Shitou goose. As far as 17 homologous sequences released by NCBI were included in analysis, 26 haplotypes were found in 53 samples, and proved that Anser cygnoides was the only ancestor for five Guangdong goose breeds based on haplotype phylogenetic analysis.
