豆类丝核菌次级代谢产物对化疗骨髓抑制小鼠的保护作用
Protective effects of secondary metabolites of Rhizoctonia leguminicola on chemotherapy induced myelosuppressive mice

摘  要: 【目的】探讨豆类丝核菌次级代谢产物对环磷酰胺所致小鼠骨髓抑制的保护作用及其作用机制。【方法】将昆明种小白鼠60只均分为6组:Ⅰ组(正常对照组)、Ⅱ组(CTX模型对照组)、Ⅲ组(苦马豆素(SW)低剂量组)、Ⅳ组(SW中剂量组)、Ⅴ组(SW高剂量组)、Ⅵ组(SW16 mg/kg)。Ⅰ、Ⅱ组灌胃生理盐水,Ⅲ、Ⅳ、Ⅴ、Ⅵ组分别灌胃浓度为8,16,32,16 mg/kg的SW液,剂量皆为0.4 mL/d,共10次;末次灌胃6 h后,Ⅱ、Ⅲ、Ⅳ、Ⅴ组腹腔注射环磷酰胺(CTX)80 mg/kg,每天1次,连续3 d,末次注射后24 h,眼眶采血并处死小鼠,测定血常规、脾脏指数、脾集落形成单位(CFU-S)、骨髓有核细胞(BMC)数及骨髓细胞增殖试验,研究豆类丝核菌次级代谢产物对小鼠骨髓造血功能的影响。【结果】腹腔注射CTX80 mg/kg连续3次,可致小鼠骨髓抑制。豆类丝核菌次级代谢产物(SW剂量为16mg/kg)可较好抑制CTX引起的白细胞数(WBC)、红细胞数(RBC)、血红蛋白含量(HgB)、骨髓有核细胞数(BMC)减少;各剂量的SW能明显增加CFU-S数,其中以SW16 mg/kg效果最佳。当SW为8 mg/kg时,能很好地刺激骨髓细胞增殖。【结论】豆类丝核菌次级代谢产物对环磷酰胺所致小鼠骨髓抑制有较好的保护作用,其机制可能是通过刺激骨髓细胞增殖,增加BMC数和髓外造血来实现。 【Objective】 The study investigated the protective effect and the mechanism of secondary metabolites of Rhizoctonia leguminicola on myelosuppression induced by cyclophosphamide in mice.【Method】 Sixty Kunming mice were randomly divided into 6 groups,GroupⅠ(normal control group),groupⅡ(CTX model control group),groupⅢ(low dose group),groupⅣ(middle dose group),groupⅤ(high dose group),groupⅥ(SW of 16 mg/kg).Group I and groupⅡwere administered with NS;groupⅢ,groupⅣ,groupⅤ,groupⅥ were administered with 8,16,32,16 mg/kg of SW;dose of O. 4 mL per day for 10 days. 6h later,group Ⅱ ,groupⅢ ,groupⅣ ,group Ⅴ were intraperitoneal injected cyclopllosphadide 8u mg/Kg, 24h after the last injection, blood routine, spleen index, spleen colony-forming units (CFU-S), bone marrow cells (BMC)and proliferation of bone marrow cells of mice were measured. [Result] Intraperitoneal injection of cyclophosphamide 80 mg/kg three times consecutly can induce bone marrow suppression. The decrease of WBC, RBC, HgB, BMC, induced by cyclophosphamide, was greatly inhibited by the secondary metabolites of Rhizoctonia leguminicola (containing SW 16 mg/kg) ;SW could significantly increase the number of CFU-S, especially when the dose of SW were 16 mg/kg; However when the dose of SW was 8 mg/kg,the proliferation of bone marrow cells of mice was greatly stimulated. [Conclusion] The secondary metabolites of Rhizoctonia leguminicola are able to protect the marrow depression induced by cyclophosphamide, and its effect is achieved probably by stimulating proliferation of bone marrow cells and increasing the number of BMC and Extramedullary hematopoiesis.
