摘 要: 为获得大白菜高效再生体系,研究了不同的6-BA和NAA浓度、供体苗态、外植体切割和接种方式对6种大白菜子叶段不定芽再生的影响。结果表明:当大白菜苗期为两片子叶绿色将展平、第一片真叶未露出、小苗直立生长时(播种5~6 d)为最佳苗态;以保留单片子叶,在生长点处(即子叶柄基部和下胚轴交接处)先横切,然后纵切,去除顶芽,切除子叶顶端1/3竖直插入培养基为最佳切割和接种方式;在MS+7.0mg/L 6-BA+0.5mg/L NAA培养基上启动培养2~3d,再转入MS+5.5mg/L 6-BA+0.5mg/LNAA芽诱导培养基培养时再生频率最高;在供试的6种基因型材料中,橙色大白菜06J28子叶段再生频率最高,为90.4%,每块外植体再生芽数最高达1.14个,这一研究结果为大白菜转基因奠定了良好基础。 In order to establish a high regeneration system of Chinese cabbage,the cotyledonary segments of six genotypes were used to study the effects of the 6-BA and NAA combinations,state of seedling,cotyledonary cutting and inoculation methods on regeneration.The results showed that the seedling growing upright with two green,lat cotyledons and no true leaf was the best stage for culture in vitro(5-6 days after sowing).The cotyledon segment was kept by cuting seedling at the cross of hypocotyls firstly, and then the apical bud, the 1/3 top of cotyledon was also cut off, lastly inserted in culture medium vertically. Cotyledon segments were induced in the medium of MS + 7.0 mg/L 6-BA + 0.5 mg/L NAA for 2-3 days, then transferred into the medium of MS + 5.5mg/L 6-BA + 0.5mg/L NAA to induce adventitious shoots, the explants of orange Chinese cabbage 06J28 showed the highest regeneration ratio, which was 90.4%, its maximum number of buds was 1.14 per explants. The results sets up a basis for transgenic improvement further.
