摘 要: 木聚糖酶是主要工业用酶制剂之一,在食品行业应用广泛。将编码枯草芽孢杆菌(Bacillus subtilis B2)木聚糖酶XYN的基因克隆到pPIC9K载体中,构建了分泌型表达载体pPIC9K-XYN,载体经线性化后转化Pichia pastoris GS115,G筛选获得了分泌表达XYN的重组毕赤酵母工程菌GS115/pPIC9K-XYN。初步研究表明:以山毛榉木聚糖为底物时,重组毕赤酵母工程菌摇瓶水平发酵上清液中木聚糖酶酶活可达1542.6U/mL,重组木聚糖酶最适温度为50℃,最适pH值为6.0,并显示出较好的热稳定性和宽pH适应性。 Xylanase is one of the important enzymes widely applied in food industries. The gene of Bacillus subtilis B2 xylanase was separated and cloned into the secreted expression vector pPIC9K to construct a recombinant plasmid pPIC9K-XYN. The plasmid was linearized and transformed into Pichiapastoris GS 115 and Pichiapastoris recombinant swain GS 115/pPIC9K-XYN was obtained. The xylanase activity of the culture supematant of recombinant strain was up to 1542 U/mL towards beenchwood xylan. The optimal temperature andpH for the recombinant xylanase was 50℃ and 6.0, respectively. Besides, the recombinant xylanase showed good thermal stability and wide adaptability to pH.
