摘 要: 目的探讨黄芪总苷对运动性疲劳大鼠胃粘膜胃泌素基因表达及氧自由基含量的影响。方法实验动物分为3组,每组8只,即正常对照组、模型组和黄芪总苷组。应用力竭游泳方法制作运动性疲劳大鼠模型,观察胃粘膜损伤指数,检测胃粘膜SOD活性、MDA含量,RT-PCR法测定胃泌素mRNA表达变化。结果黄芪总苷预防给药可以明显减轻运动性疲劳大鼠胃粘膜损伤指数,降低MDA含量,升高SOD活性,显著增加胃泌素基因表达。结论黄芪总苷对运动性疲劳大鼠胃粘膜损伤有保护作用,其保护机制可能与增加胃泌素基因表达,并参与抗氧自由基损伤有关。 Objective To investigate the effects of aswagaloside (AST) on gastrin gene expression and the free radical content of rots with exercise fatigue. Methods The experimental animals were divided into 3 group, such as the normal group, the model group, and the AST group. The model of rats with exercise fatigue was produced by the method of exhausting swimming exercise. The gastric mucosal ulcer index, SOD activity, MDA content were observed, the expression of gastrin mRNA were detected in gastric mucesa by RT- PCR. Results Intragastric administration of AST markedly decreased the ulcer index and the MDA content, and significantly increased the SOD activity and the gastrin gene expression in gastrin mucosa of model rat. Conclusion AST may protect gastric mucosa from ulcer lesion by exhausting exercise, the mechanism may be related to AST raise the gastrin gene expression and against free radical damage.
