摘 要: 运用紫外吸收光谱、荧光光谱和CD光谱研究氧化异阿朴菲生物碱衍生物——9-(3-吡咯基丙酰胺基)-1-氮杂苯并蒽酮与DNA的相互作用。研究表明化合物的大环共平面体系可能嵌入到双螺旋DNA的碱基对中,且优先结合到双螺旋DNA的AT序列上。CD光谱研究确定了化合物在与DNA结合时,其生色团的长轴与DNA碱基对的长轴平行。 The interaction of oxoisoaporphine derivative 9-[-3-Pyrrolidinopropionamido]-l-azabenzan-throne with calf thymus DNA (ct DNA),Poly(dAT)z and Poly(dGC)2 has been studied by fluorescence, CD and UV spectra. A strong electronic interaction between the oxoisoaporphine moiety and the DNA bases was found by UV spectra. Experiment of ethidium displacement indicated that derivative was in favor of binding on AT sequence. The induced circular dichroism (ICD) indicated that the long axis of the derivative was oriented parallel to the base pair long axis.
