提高漆酶活力的菌株组合筛选及诱导剂的研究

摘  要: 采用木质素降解酶互补的产酶组合培养和碱溶木质素降解相结合的方法,对实验室保存和分离的9株木质素降解菌株进行组合培养实验。结果表明,编号为m-6、55、m-8三株菌组合时,提高了整体产漆酶量,最高酶活达到了891.7 U/L,比单菌株m-6的产酶量提高了6倍。该组合菌的最适产酶pH值为5.5,最适产酶温度为32℃。在对其产酶特性的研究中发现,外加藜芦醇5 mmol/L、吐温-80 0.01%和愈创木酚3 mmol/L,对组合菌株分泌漆酶有很大的促进作用,漆酶最高酶活达到2152.8 U/L,比对照样提高了166.3%。固态产酶培养后,第30天时,稻草粉的失重可达51.4%,纤维素降解率为38.5%,木质素的降解率为45.2%。 The combined strains, named m-6, 55, m-8 were screened from 9 lignin degradation fungi reserved by our lab in the way of mixed cultivating in liquid medium for secreting lignin degradation enzyme and alkali soluble lignin degradation. It showed that the maximum lacca- se enzyme activity of combined strains is 891.7 U/L, which is 6 times of m-6 alone. The optimum temperature and pH for the combined strains is 32℃ and 5.5 respectively. When Veratryl alcohol 5 retool/L, Guaiacol 3 mmol/L and Tween-80 0.01% were added in liquid medium, they promoted laccase secreted greatly. The maximum laccase enzyme activity of the combined strains is 2152.8 U/L, which is increased 166.3% compared with the control. After 30 days cultivation in solid medium, the weight of straw reduces 51.4%, the lignin degradation of straw is 45.2% and the cellulose degradation of straw is 38.5%.
