摘 要: 通过热水浸提法从荞麦中提取水溶性多糖,并用离子交换柱和凝胶过滤柱对荞麦多糖进行分离纯化、测定其分子量。结果表明,荞麦多糖粗品用Savag法脱蛋白后,采用DEAE-Sepharose Fast Flow和Sephacryl S-400HR柱层析纯化,得到荞麦多糖(FEP),经过凝胶柱色谱分析表明,FEP为均一多糖。用苯酚硫酸法测定多糖的含量,用AKTA快速液相蛋白层析仪的紫外检测器在280nm波长处检测蛋白质含量。最后根据用不同分子量标准葡聚糖做出的标准曲线,测定出FEP分子量(Mw)为1720442D。 A water-soluble polysaccharide from Fagopyum esculentum (FEP) was obtained by boiling water extraction and ethanol precipitation, followed by consecutive purification on DEAE-Sepharose Fast Flow column and Sephacryl S-400 HR column, and was subjected to freeze-drying. The elution profile of FEP on Sephacryl S-400 HR column presented only a single symmetrical peak, indicating that the FEP was purified to a homogeneous polysaccharide. The polysaccharide content in dried FEP powder was 98.5% determined by phenol-sulfuric acid assay, and no protein components were detected in the powder. The molecular weight of the FEP was calculated as 1720442 D, according to the calibration curve of standard dextrans.
