康宁木霉与米根霉混合发酵生产纤维素酶和木聚糖酶的研究

摘  要: 对康宁木霉QF-02和米根霉NRRL395液态混合发酵生产纤维素酶和木聚糖酶的工艺进行了研究.实验结果表明:康宁木霉先接种培养48 h后再接种米根霉产酶效果最佳,在此接种方式下获得的优化培养温度为28℃,起始pH为5.0.混合菌在发酵罐中的产酶趋势与康宁木霉纯培养类似,最大酶活都出现在120 h,但混合培养物中FPA、β-葡萄糖苷酶活和木聚糖酶活比纯培养分别提高14.8%、45.4%和4.3%.米根霉的加入使混合培养物中还原糖浓度下降并维持在较低水平,而康宁木霉纯培养中还原糖呈不断增加的趋势.康宁木霉和米根霉混合发酵产酶的微生态原理应归于二者形成了相互促进的共生关系. The production process of cellulase and xylanase through mixed fermention of Trichoderma koningii QF -02 and Rhizopus oryzae NRRL395 was investigated. The results show that optimal enzymatic activities can be achieved by inoculating R. oryzae after inoculating and culturing T. koningii for 48 h, and the optimal culture temperature and the initial pH value were 28 ℃and 5.0 respectively in this inoculation mode. The enzyme production tendency of the mixed bacteria in the fermentor is similar to that of pure culture of T. koningii, and the maximal enzymatic activities occur at 120 h in both cases, but FPA and the activities of β-glucosidase and xylanase in the mixed culture are respectively enhanced by 14.8% , 45.4% and 4.3% in comparison with pure culture. The addition of R. oryzae can lower the concentration of reducing sugar in mixed culture and keep it at a lower level, whereas reducing sugar in the pure culture of T. koningii increases continuously. The microecological principle for mixed fermentation of T. koningii and R. oryzae attribute to their favorable symbiotic relationship.
