摘 要: 采用酶解技术制备具ACE抑制活性的大豆肽混合物。选择了最佳水解用酶,优化了反应条件。确定碱性蛋白酶解最佳条件为:pH9.0,温度50℃,底物浓度6%,酶用量([E]/[S])3%,作用时间4h。选择D3520型大孔树脂对水解产物进行脱盐精制,脱盐率91.2%,肽回收率89.3%,ACE抑制活性提高了28.5%,大豆分离蛋白ACE抑制肽混合物分子质量主要分布为200-720,在pH3-10,溶解性达98.1%以上,且稳定性好。 SPI were hydrolysated by enzyme catalysis technique to prepare angiotensin converting enzyme inhibitory peptide. Orthogonal design of four factors three levels was taken to optimize the hydrolysis conditions of SPI with Alcalase protease. The results showed that the best condition was: pH9. 0, temperature 50℃, enzyme concentration 3%, substrate concentration 6%, reaction time 4h. D3520 macro porous adsorbed resin were utilized to adsorb ACE Inhibitory Peptides from SPI, and the content of ash of ACE Inhibitory Peptides were 8.8%, recovery rate of ACE Inhibitory Peptides more than 89.3%. The relative molecular weight of ACE Inhibitory Peptides from SPI mainly was distributed from 200 to 720, and its solubility was about 98.1% with the pH value from 3 to 10.
