摘 要: 采用禽白血病J亚群抗体检测试剂盒对4个不同肉种鸡场的血清样本进行检测,发现ALV-J抗体阳性率平均达到16.52%(3.33%~30.0%)。对出现肿瘤病变的显性型病鸡进行病理组织学检查,在肝、肾、脾组织中观察到大量增生的、含嗜酸性颗粒的髓细胞样瘤细胞。针对J亚群gp85基因部分序列进行PCR扩增,从ALV-J抗体阳性鸡的肝病料扩增出一条545bp DNA带,而ALV-J抗体阴性鸡的样本显现阴性反应。将扩增的gp85基因序列与国内外J亚群毒株相应序列进行比较,结果核苷酸序列的同源性分别达到94.8%~96.5%和95.5%~96.8%。综合血清学、病理组织学变化和病原分子生物学鉴定结果,证实贵州省肉种鸡群中存在ALV-J亚群病毒感染。 The serum samples were collected from 4 different breeding broiler farms and detected with avian leukosis virus antiboby test kit-subgroup J (ALV-J). The positive rate of ALV-J antibody averaged 16.52% in different broiler flocks (3.33%-30,0%).The histopathological examination for the sick or dead chickens which manifested tumor lesions in visceral organs showed a lot of myelocyte-like tumor cells filled with acidphilus granules in liver,kidney and spleen tissues, gp85 gene of subgroup-J virus was amplified by PCR and a 545 bp fragment was amplified from livers of chickens which were positive to ALV-J antibody. No fragment was observed from samples of ALV-J antibody negative chickens. Compared with the relative sequence of subgroup J isolates published in GenBank,the homology of nucleotide sequences reached 94.8%-96.5% and 95.5%-96.8% respectively. These results demonstrated ALV-J infection in breeding broiler flocks of Guizhou province.
