摘 要: [目的]在对OsWNK基因的生化特性有了一定了解的基础上,研究OsWNK在水稻根组织的表达,为进一步研究基因OsWNK的功能奠定基础。[方法]利用RNA原位杂交技术,通过制备OsWNK基因的正反义地高辛标记的RNA探针,与水稻根内靶基因转录表达产物进行原位杂交定位。[结果]在根尖生长点部位的OsWNK表达最旺盛,而在伸长区的细胞中,OsWNK的表达量很低。另外,在根冠中几乎没有OsWNK的表达。[结论]OsWNK在根尖生长点的大量分布说明OsWNK在植物快速生长的组织和细胞中表达比较高,推测此基因可能与生长点细胞频繁的细胞分裂有关。 [ Objective ] On the basis of understanding of the biochemical performance of OsWNK, its expression pattern in rice root tissue was explored to lay the foundation for the research on OsWNK function. [ Method] With in situ RNA hybridization technique, the sense and anti - sense OsWNK probes were hybridized with the products expressed by the target gene in rice root tissue. [ Results] The experimental results showed that OsWNK had a high expression in the growing point cells of root tip, while its expression was very weak in elongate area cells and there was no expression detected in root cap ceils. [ Conclusion] OsWNK was mainly accumulated in the fast growth cells and tissues such as growing point cells of root tip, which meant that OsWNK might play roles in cell division.
