转鲑鱼降钙素基因酵母的亚急性毒性实验

摘  要: 评价转鲑鱼降钙素基因酵母的安全性。取转基因酵母分别以高(5 g/kg)、中(0.5 g/kg)、低(0.1 g/kg)3种剂量组对大鼠连续灌胃给药,30 d后观察其行为,测试大鼠体重、血常规、生化指标及病理组织学观察。结果:转基因酵母各剂量组动物的一般状况、体重、主要脏器系数、血液学指标及血液生化指标,与正常对照组比较,均未见明显差异;病理检查未见与药物毒性相关的主要脏器组织形态学改变。转鲑鱼降钙素基因酵母对实验大鼠安全无毒,且在试验期内本试验所用剂量对于试验大鼠来说是安全的。 Objective:to evaluate the toxicological safety of recombinant sacchromyces cerevisiae expressing salmon calcitonin in order to provide some data for its application.Methods: rats in 3 experimental groups were treated orally with saccharomyces cerevisiae at the dosages of 5 g/kg BW,0.5 g/kg BW and 0.1 g/kg BW successivlly for one month.Then observed the sign and behavior,determined the blood routine,blood biochemical index and examined the pathologic index of visceral organs of rats.Results: In the test,the results showed that the diet,weight,blood routine examination,blood biochemical criterion and major organ index of rats were normal.Major organs were not found abnormial in pathological test.Over all there were no significant differences in the study groups compared with the control group.Conclusion: No acute toxicity is found with saccharomyces cerevisiae,and it may be safe to be used in clinic.
