大鼠脑缺血再灌注损伤后皮层Homerla蛋白改变及意义

摘  要: 目的探讨大鼠脑血再灌注损伤后皮层组织Homer1a蛋白的表达改变及其意义。方法选择成年雄性SD大鼠59只,随机分为正常对照组、缺血再灌注损伤后10min、1h、3h、6h、12h、24h、48h及72h共9组,每组6只。免疫组化染色法和Western blot法测定Homer1a蛋白含量,T-PCR法测定Homer1aRNA表达。结果在实验过程中5只动物死亡,54只进入结果分析。与对照组比较缺血再灌注损伤后Homer1a蛋白和mRNA灰度值表达出现3个峰值,分别在10min、6h和24h,但在72h仍维持较高水平;与对照组Homer1a蛋白免疫评分相比,缺血再灌注损伤后出现两个峰值,分别在6h和24h;Homer1a蛋白除神经元外,在胶质细胞也表达。结论在缺血再灌注损伤后,Homer1a出现3个表达高峰,可能在不同的机制的影响下对脑损伤发生发展起重要作用。 To investigate the expression and significance of Homerla protein in cerebral cortexafter ischemic reperfusion injury of brain in rats. Methods Fifty-four adult SD rats were randomized into nine groups with 6 rats in each group : normal control group and post-ischemic groups of 10 min, 1 h, 3 h, 6 h, 12 h, 24 h,48 h and 72 h after ischemic brain injury. The expressions of Homerl a were investigated by immunohistochemistry, immunoblotting and RT-PCR. Results Forty-nine rats were studied with five rats excluded due to their death during the study. It was shown that compared with the control group, there were three peak levels of expression of the gray value of Homerla protein and mRNA following ischemic reperfusion brain injury, at 10 min, 6 h and 24 h. The levels remains high at 72 h with mild decrease. The immunoreactivity score of Homerla protein expression showed two peak values respectively at 6 h and 24 h after ischemic reperfusion brain injury; Homerl a protein expression was also detected in glia cells besides neurons. Conclusions After ischemic reperfusion brain injury, there are three peak levels of expression of Homerl a which may play important roles in the development of brain injury via different mechanism.
