摘 要: 研究Sca-1(stemcell antigen-1,干细胞抗原),CD24和Muc1(Mucin1)在雌性大鼠乳腺中的分布。制备6周、9周、孕15 d、产后3 d SD(Sprague-Dawley)雌性大鼠乳腺标本切片(4μm),运用免疫组化酶标法对乳腺中Sca-1,CD24,Muc1进行单标记;用Western免疫印迹法检测Sca-1,CD24及Muc1在6周、9周、孕15 d、产后3 d大鼠乳腺中的含量变化。结果表明:Sca-1、CD24、Muc1在6周、9周、孕15 d、产后3 d SD雌性大鼠乳腺中不同部位有不同程度的分布。乳腺祖细胞标记物Sca-1在乳腺静止期的6周、9周分布较多,孕15 d以及产后3 d分布减少,提示一部分细胞转化为成熟的乳腺细胞;乳腺干细胞标记物CD24在乳腺发育的静止期及孕期数量较为稳定;成熟乳腺上皮细胞Muc1在乳腺静止期的6周、9周分布较少,孕期及产后随着乳腺的发育,乳腺上皮细胞的增殖,成熟乳腺上皮细胞逐渐增多。 To study the distribution of Sca- 1, CD24 and Mucl in mammary glands of female rats. We prepared 6 week, 9 week, gravidity 15 d, 3 d of postnatal female mammary gland slice (4μm), and detected Sca-1, CD24 and Mucl of mammary gland by immunohistochemical enzyme labeling. The content of Sca-1 ,CD24 and Mucl during 6 week, 9 week, 15 d of gestation, 3 d of postnatal of female nmmnmry gland were executed by Western blotting. Results showed that Sca-1, CD24,Mucl distributed in different degree and different position in SD'S mammary gland of 6 week,9 week, 15 d of gestation,3 d of postnatal.As a mammary gland progenitor cell marker, the content of Sca- 1 does not change obviously during 6 week, 9 week, 15 d of gestation, but decreases in 3 d of postnatal, it means that part of mammary gland cells transformes to ripe ones. As a mammary gland stem cell marker, the content of CD24 is stable during mammogenesis quiescence and active stages, as a maturemammary gland epithelium cell marker, the distribution of Mucl in 6 week,9 week mammary gland are less than that in pregnancy and puerperium, epithelial cells vegetating accompany mammary gland developing, mature mammary gland epithelial ceils increase gradually.
