山羊内毒素休克时山莨菪碱对肾脏自由基代谢的影响
Effects of anisodamine on renal free radical metabolism during endotoxic shock of goats

摘  要: 36头杂交山羊随机分成3组,第Ⅰ组静脉注射生理盐水作为对照;第Ⅱ组静脉注射大肠杆菌(O111B4)内毒素,1800U/kg,注射两次,间隔24h;第Ⅲ组处理同第Ⅱ组,但于第2次注射内毒素前10min静脉注射山莨菪碱2.5mg/kg。分别于试验后第5和12h各组剖杀6头,检测肾组织中丙二醛(MDA)含量和抗氧化参数。结果表明:第Ⅱ组肾组织MDA含量明显升高(P<0.01),谷胱甘肽过氧化物酶(GSH-Px)、谷胱甘肽还原酶(GR)、总超氧化物歧化酶(T-SOD)、锰-SOD、铜锌-SOD活性均显著下降(P<0.05),并伴有还原型谷胱甘肽(GSH)、总巯基(T-SH)、非蛋白巯基(NP-SH)、蛋白结合巯基(PB-SH)含量明显降低(P<0.01,P<0.05);第Ⅲ组肾组织MDA含量在试验后5h低于Ⅱ组(P<0.05),试验后12h与Ⅰ组差异不显著(P>0.05),GSH-Px、GR、CuZn-SOD和试验后5h的T-SOD、Mn-SOD活性均高于Ⅱ组(P<0.05),GSH、T-SH、NP-SH、PB-SH含量亦高于Ⅱ组(P<0.05),其中T-SH、PB-SH与Ⅰ组无明显差异。提示在山羊内毒素休克时,肾? Thirty six hybrid goats were randomly divided into three groups. Group Ⅰ was the normal control. The goats of Group Ⅱ were intravenously injected E. coli (O 111 B 4) endotoxin with dose of 1 800 endotoxin unit per kilogram body weight for 24 h apart. The goats of Group Ⅲ were treated with the same as Group Ⅱ and intravenously given 2.5 mg anisodamine per kilogram body weight for 10 minutes before the second endotoxin administration. Six goats of every group were killed 5 h and 12 h after endotoxin injection. Renal tissue MDA contents and antioxidant parameters were examined respectively. The results showed as follows. The renal tissue MDA contents of Group Ⅱ significantly increased ( P <0.01). The activities of renal GSH Px, GR, T SOD, Mn SOD and CuZn SOD markedly decreased followed with reduction of the levels of GSH, T SH, NP SH and PB SH of renal tissues in Group Ⅱ. However, the renal tissue MDA contents of Group Ⅲ were lower than those of Group Ⅱ at 5 h and not different with Group Ⅰ at 12 h. The activities of GSH Px, GR, CuZn SOD and T SOD, Mn SOD of Group Ⅲ at 5 h were higher than those of Group Ⅱ ( P <0.05). The contents of renal tissue GSH, T SH, NP SH and PB SH of Group Ⅲ were more than those of Group Ⅱ. There were no difference of renal tissue T SH and PB SH between Group Ⅰ and Ⅲ. The experimental evidence indicated that renal antioxidant function significantly decreased and renal lipid peroxidation injury induced by free radical during endotoxic shock of goats, but anisodamine given before endotoxin administration could markedly reduce the renal tissue injury.
