摘 要: 以中棉35号、中棉27、石远321和珂字棉312共4个陆地棉(Gossypium hirsutum L.)品种茎尖为材料,探讨了苗龄、生长调节剂、活性炭和IBA等因子对茎尖培养和再生的影响。结果表明,5d苗龄的茎尖、0.1mg/L6-BA+0.1mg/Lα-NAA最适于不定芽的诱导,3g/L活性炭和0.4mg/LIBA对根诱导与植株再生效果最佳,再生频率平均可达86.8%。实验结果初步建立了棉花茎尖的再生体系,为棉花茎尖的遗传转化奠定了基础。 Shoot tips of Chinese elite cotton (Gossypium hirsutum L.) varieties Zhongmian 35,Zhongmian 27,Shiyuan 321 and Coker 312 were excised from the seedlings and cultured on the MSB medium.Effects of seedling age,hormonal regimes,activated charcoal (AC) and IBA on regeneration of shoots were studied.The results showed that 5-day seedling,0.1 mg/L 6-BA and 0.1 mg/L α-NAA were suitable for induction of multiple tips,3 g/L AC and 0.4 mg/L IBA had optimal effects on the rate of plantlet regeneration,which averaged 86.8%.At last,a stable shoots regeneration system of cotton for further genetic transformation was established.
