摘 要: 采用PCR技术对甘肃省境内4种鼢鼠线粒体DNA控制区序列进行扩增测序,获得了23条长度为414~424 bp的同源序列.通过序列比较分析,共检出179个多态信息位点,定义了15种单倍型,单倍型多样度(Hd)为:0.941±0.340.同源基因序列平均含AT碱基65.6%,GC碱基34.4%.基于控制区对其构建的分子进化树表明:4种鼢鼠构成2个进化枝,高原鼢鼠和斯氏鼢鼠先聚在一起,后与秦岭鼢鼠聚在一起构成一枝;甘肃鼢鼠构成另一枝,处于基部,其分别与采集的三个地理种群相对应. To evaluate systematical evolution relationships, the mitochondrial control region of four kinds of zokors were investigated by PCR and direct sequencing. 23 individuals of 414-424 bp homologous nucleotide sequences were obtained. The results showed 179 polymorphic nueleotide sites among homologous sequence,and 15 haplotypes were found and the AT,GC content was 65.6 %,34.4 based on the control region showed that two in all individuals. The haplotype diversity was 0. 941±0. 340, % respectively. In addition, the molecular phylogenetic trees major evolutionary lineages were firstly recognized within the specimens,Clade A(M. baileyi,M, smithi and M. ru fecens) and Clade B (M. cansus) ,while M. cansus remained the basal elade among the four species and the three clades corresponding to the major geographical populations.
