黄皮和杨梅果肉中绿原酸、表儿茶素及芦丁含量的HPLC测定
Determination of Chlorogenic Acid,Epicatechin and Rutin in Wampee and Wax Myrtle Flesh by HPLC

摘  要: 应用高效液相色谱(HPLC)法测定了黄皮、杨梅果肉中的绿原酸、表儿茶素和芦丁的含量。色谱柱为Nucleosil C18反相柱(250 mm×4 mm),流动相为20%的甲醇(A),30%乙腈(B),pH 3.2,梯度洗脱,流速0.8mL/min,检测波长280 nm,柱温35℃。结果表明:3种标准品的线性范围为0.390 6~50.000 0μg/mL,相关系数r=0.999 9。标准品在0~48 h内经多次测定,保留时间和峰面积的相对标准偏差分别为0.78%~0.91%和0.67%~0.95%;回收率和回收率相对标准偏差分别为93.21%~103.50%和2.77%~3.94%;仪器最低检出限分别为0.090 1、0.095 5、0.098 0μg/mL。测定结果还表明:黄皮和杨梅果肉中均含有一定量的绿原酸、表儿茶素和芦丁。黄皮的4个品种中,3种物质的含量(鲜重)分别为4.046 8~38.669 0、4.316 4~84.614 8、52.392 4~130.609 7μg/g;杨梅的4个品种含量(鲜重)分别为17.564 4~93.546 3、113.090 8~236.401 5、46.366 7~168.370 0μg/g。 A high-performance liquid chromatography(HPLC) method to analyze chrologenic acid, epicatechin and rutin in wampee and wax myrtle flesh has been developed. Samples were injected into reversed-phase Nucleosil C18 column(250 mm× 4 mm). Mobile phase A was 20%0 methanol and mobile phase B was 30% acetonitrile, pH was adjusted to 3. 2. Gradient elution proceeded at the flow rate 0. 8 mL/min. Operation conditions were as follows, column temperature 35 ℃, detection at 280 nm. The results showed that: Linear calibration of three standard samples were within 0. 390 6-50. 000 0 μg/mL with correlation 0. 999 9. Relative standard deviation(RSD) of run time and peak area were respectively within 0. 78% -0. 91 % and 0. 67 -0. 95 % after standard samples were repeated analyzed in 48 h. Recovery was within 93.21%-103. 50% with RSD of 2. 77%-3. 94%. The limit of detection (LOD) were 0. 090 1, 0. 095 5, 0. 098 04μg/mL. The method was validated for chrologenic acid, epicatechin and rutin present in flesh of wampee and wax myrtle. The contents of those three substances were within 4. 046 8-38. 669 0, 4. 316 4-84. 614 8 , 52. 392 4-130. 609 7μg/g fresh weight in four wampee varieties, and within 17. 564 4-93. 546 3, 113. 090 8-236. 401 5, 46. 366 7-168. 370 0 μg/g fresh weight in four wax myrtle varieties.
