摘 要: 基于Berthelot显色反应,研究L-谷氨酸在谷氨酸脱羧酶催化下产物γ-氨基丁酸含量测定的比色法。实验确定测定γ-氨基丁酸适宜反应条件为酶反应液1.0 mL,1.0 mol/L Na2CO3溶液0.2 mL,0.01 mol/L四硼酸钠缓冲液1.0 mL,6%苯酚溶液1.0 mL,7.5%NaClO溶液5.0 mL,沸水浴10 min,冰浴5 min,60%乙醇溶液2.0 mL,测定OD640绘制标准曲线并计算样品中γ-氨基丁酸的含量。结果表明,该方法灵敏度较高、重现性较好、测量相对误差<5%、操作简单易行、设备要求简单,适合大批量样品的快速分析。 With the principle of chromogenic reaction, the paper studied the colorimetric method to determine y-aminobutyric acid produced from glutamate in the presence of L-Glutamate decarboxylase. The optimum conditions for the determination of γ-aminobutyric acid were: enzymatic reaction liquid lml, Na2CO3(moVL)0.2 mL, sodium tetraborate (0.01 mol/L, pH9.18) 1 mL, 6% phenol 1 mL and then add 7.5% NaCIO 5 mL. Placed the liquid in the boiling water for 10min and then in the ice for 5min. Added 2.0 mL 60% Alcohol to the liquid and determined at 640 nm. The content of y-aminobutyric acid in the sample could be calculated with the contrast of standard substance. The result showed that, the colorimetric method had the advantages of high sensitivity, reappearance with the relative error〈5%. It had low demand for the equipment and was operated simply. The colorimetric method was suitable for quick analysis of amount of samples but had to remove the dissociated amino acid.
