摘 要: 通过对截获于澳门入境旅客携带芒果中的实蝇(SH78)进行线粒体COⅠ、COⅡ和COⅢ基因序列的扩增测序,并与GenBank中相关的序列进行比对,结果表明截获实蝇样品线粒体COⅠ基因序列和柱果小条实蝇Ceratitis cosyra(AY788421)相似性为98.87%;COⅡ基因序列与柱果小条实蝇(C.cosyra) (AY805310)和C.cosyra(AY805311)相似性分别为99.27%和98.11%;COⅢ基因序列和C.cosyra (DQ462338)相似性为89.10%。基于COⅠ和COⅡ序列构建的系统发育树中,截获实蝇样品和杧果小条实蝇最为接近。根据序列分析和系统发育关系分析的结果,将截获的实蝇鉴定为杧果小条实蝇(Ceratitis cosyra)。鉴定结果与形态鉴定结果相同。 The mitochondrial gene, including the mitochondrial cytochrome oxidase subunit Ⅰ (CO Ⅰ ) gene, CO Ⅱ gene and CO Ⅲ gene, of the fly larvae SH78 were intercepted in mango carried in Shanghai by Macao flight, were amplified and sequenced with primers designed from Ceratitis capitata (AF189691, AJ242872) and Bactrocera oleae (AY210702, AY210703). The CO Ⅰ , CO Ⅱand CO m gene sequence were obtained and analyzed with the homologous sequence of related species in GenBank. The result showed that the similarity of CO Ⅰ gene sequence are 98.87% between SH78 and C. cosyra ( AY788421 ). The similarity of CO Ⅱ gene sequence are 99.27% ( AY805310), 98.11% ( AY805311 ), respectively. CO Ⅲ gene sequence with the similarity of 90. 00% between SH78 and C. cosyra (DQ462338). The molecular phylogenetic relationship of 15 species of Tephritidae based on CO Ⅰ and CO Ⅱ gene sequences showed that SH78 and C. cosyra (AY805310) are within the same subclade. The larvae(SH78) was identified as C. cosyra with the analysis of sequence and phylogenetic relationship.
