摘　　要： 分别以甜瓜品种"东方蜜1号"的成熟干种子、萌动种子、子叶和真叶为材料,选用4种不同的提取方法进行总DNA提取试验。结果表明:包括干种子在内的不同生长期的材料均能提取到总DNA,刚展平子叶的提取效果最好;4种提取方法中以尿素提取法最为简便,效果最佳。经PCR检验,干种子和子叶提取的总DNA均可用于PCR扩增。 The tests for extracting the total DNA respectively from the dry seeds,germinated seeds,cotyledons and true leaves of the melon cultivar ＂Dongfangmi 1＂ were carried out by four different methods.Results showed that the total DNA could be extracted from all the materials,but the quality of the DNA extracted from flattened cotyledons was the best.Among four different methods,the urea method was the most convenient and effective.The PCR test showed that the total DNAs extracted from the seeds and cotyledons could be both used for PCR amplification.
