ELISA,Dot-blot and RT-PCR were developed and evaluated for detection of the virus in diseased banana plants.RT-PCR was the most sensitive assay when compared with Dot-blot and ELISA.The Dot-blot assay was 100 times more sensitive than ELISA.The distribution of CMV within banana plants was uneven,CMV concentritions were higher in younger leaves than in older ones,as well as higher in the midribs than the leaves.The CMV-Oahu isolate was generally not detected in banana by ELISA using antibodies to both CMV subgroups Ⅰ and Ⅱ,but was positive in dot-blot and RT-PCR tests.
