摘 要: 应用HPLC法分析了黄皮和杨梅果肉中核苷的含量.色谱柱为Nucleosil C18反相柱(250mm×4.6mm,5μm),流动相为水和甲醇,二元梯度洗脱,流速0.8mL/min;柱温40℃;分析波长为250nm.结果表明,黄皮4个品种果肉中含次黄嘌呤核苷16.5682-40.6261μg/g鲜质量、腺苷类物质16.2405-39.5125μg/g鲜质量;杨梅4个品种果肉中含次黄嘌呤核苷1.4038-9.0940μg/g鲜质量、腺苷类物质8.0943-20.0012μg/g鲜质量,另外还含有胸苷类物质6.1806-61.9100μg/g鲜质量. A method of high-performance liquid chromatography (HPLC) for analyzing nucleosides in the flesh of wampee and waxberry was developed. Samples were injected into reversed-phase Nucleosil C18 column (250 mm × 4. 6 mm, 5 μm). Water and methanol were used as dual gradient elution at the flow rate 0. 8 mL/min. Operation conditions were set as follows: 40℃ of column temperature, 260 nm of detection wavelengh. The method was validated for inosine and adenosine present in the flesh of wampee and waxberry. Thymidine was also detected in the flesh of waxberry. Contents of inosine and adenosine were respectively 16.568 2-40.626 1 μg/g fresh mass (FM) and 16.240 5 -39.512 5 μg/g FM, respectively, in the flesh of four wampee varieties. Contents of inosine, adenosine and thymidine were 1.403 8 - 9. 094 0 μg/g FM, 16. 240 5 -39. 512 5 μg/g FM and 6. 180 6 -61.910 0 μg/g FM, respectively, in the flesh of four waxberry varieties.
